To characterize the caspase-9 signaling cascade in intact cells, we employed chemically induced dimerization to activate caspase-9 specifically. Dimerization of caspase-9 led to rapid activation of effector caspases, including caspases-3, -6 and -7, as well as initiator caspases, including caspases-2, -8 and -10, in H9 and Jurkat cells.

These communications between cells triggers intracellular signaling cascades, termed signal transduction pathways, that regulate specific cellular functions. Each signal transduction occurs with a primary extracellular messenger that binds to a transmembrane or nuclear receptor, initiating intracellular signals. The effectiveness of the intracellular regulation is affected strongly by the characteristics of signal propagation through the cascade. For signals that are amplified as they propagate through the cascade, reactions in the upstream levels of the cascade exhibit much larger sensitivities to regulation by release of phosphorylated substrate protein and kinase than downstream reactions.

Lactate signaling cascade was recently linked to some cardiovascular disorders for its regulatory functions to myocardial structural remodeling.

Intracellular signaling cascade involves many components, and their modifications occur by enzymes. Phosphorylation is the most common chemical modification that takes place during intracellular signaling; it activates enzymes that are essential for the downstream process. Moreover, phosphorylation causes changes in their shapes.

Aspirin, for example, acts by blocking a specific lipid signalling pathway. In intracellular signalling a signal arriving at the cell surface is received by a receptor

Introduction:Intracellular signaling cascades including mTOR (mammalian target of rapamycin), MEK (mitogen-activated protein kinase), BCL2 (B-cell lym Intracellular signaling pathways downstream from glutamatergic signaling and regulation by serotonin, which lead to communication between pre- and postsynaptic elements, leading to distinct neural patterns in the barrel cortex, are emerging (reviewed in Barnett et al., 2006a; Rebsam and Gaspar, 2006). Intracellular signaling is one of the most widely studied areas in biology. Extracellular information perceived at the surface of a cell must be translated into an intracellular response that involves a complex network of interwoven signaling cascades.

We derive a reaction-diffusion model of the mitogen-activated protein kinase cascade, and use it to show how oscillations of the protein kinase concentrations can occur as a function of the depth of the cascade. In living organisms, cascades of covalent modification cycles are one of the major intracellular signaling mechanisms, allowing to transduce physical or chemical stimuli of the external world into Intracellular signaling cascades amplify a signal originating extracellularly and directs it to its intended intracellular target resulting in transcription, translation, protein modifications, enzyme activation, cellular metabolism, mitosis, and/or apoptosis. The most basic of signaling cascades involves the activation of second messengers and the Since all-or-none decisions of the cell are controlled by extracellular signals, cells have biochemical switches within their intracellular signaling networks. Central elements of these switches are multisite phosphorylation, enzymic saturation, and amplification by cascades. These communications between cells triggers intracellular signaling cascades, termed signal transduction pathways, that regulate specific cellular functions.

Light, odorants and taste molecules operate through a signal cascade that includes activation of adenylate (AC) and guanylate cyclase (GC), and subsequent opening or closing of cyclic-nucleotide-gated channels whose gating is regulated by the cyclic nucletotides cAMP or cGMP. Intracellular signaling cascade Primary Antibodies: Thermo Fisher antibodies are validated for applications including western blotting, immunocytochemistry, flow cytometry, and chromatin immunoprecipitation. The changes elicited by ligand binding (or signal sensing) in a receptor give rise to a biochemical cascade, which is a chain of biochemical events known as a signaling pathway. When signaling pathways interact with one another they form networks, which allow cellular responses to be coordinated, often by combinatorial signaling events.
