Using a recently-described, fully-in vitro system we selected synthetic CD72-specific nanobodies, incorporated them into CARs, and demonstrated robust activity against B-cell malignancy models, including CD19 loss. We have developed a direct proteomic approach to explore the cell-surface proteome for all four embryo-derived stem cell lines using affinity labeling and mass spectrometry. Antibodies against lineage-specific cell-surface proteins enabled identification and isolation of specific cell populations during stem cell differentiation and reprogramming. This review will introduce proteomics technology and outline approaches for enriching for cell surface proteins. The mammalian sperm cell at fertilization will be used as an exemplar and we argue that enriching the plasma membrane is critical to map proteins on the cell surface in order to further understand early fertilization events (capacitation, zona binding and acrosomal exocytosis). 2020-05-12 · Cell-Surface Proteomics of Naive and Primed Human Pluripotent Stem Cells (A) Plasma membrane profiling and mass spectrometry to quantify cell-surface protein expression in hPSC.

Proteolytic cleavage events at the cell surface often evade detection by conventional proteomics methods owing to their low relative abundance compared to cytoskeletal and cytoplasmic proteins.
The proteome is the entire set of proteins that is produced or modified by an organism or system. Proteomics has enabled the identification of ever increasing numbers of protein. This varies with time and distinct requirements, or stresses, that a cell or organism undergoes

Cell surface proteomics is the study of cell surface proteins, which can be essential in understanding protein structure, function, modifications they undergo, and more. This information can be crucial in identifying potential biomarkers of disease, such as cancer, and can be influential in the establishment of innovative therapies.
The knowledge of the transient changes on the surfaceome upon a perturbation is very important as the initial changed proteins could be driving molecules for some phenotype.
